Corning® Nonenzymatic Cell Dissociation Agents
By CorningĀ®Cellstripper® is a non-enzymatic cell dissociation solution formulated with a proprietary mixture of chelators which gently dislodges adherent cells in tissue culture. It is designed to be an alternative to trypsin when used in conjunction with serum-free or serum-containing media. Cells can be exposed to Cellstripper® for longer periods of time without the risk of damage associated with protein digestive enzymes like trypsin. Experimental comparisons of dissociation solutions for cell detachment reveal that Cellstripper® achieves superior results in comparison to enzymatic dissociation products.
The following instructions are applicable to most cell lines. Actual procedures and concentrations should be determined by experience with individual cell lines. Researchers should regularly monitor cell viability at sub-culturing to determine the procedures most suitable to culture conditions.
Instructions for Use
- Pre-warm Cellstripper® to incubator temperature. This can be accomplished using a 37°C water bath or by simply placing the bottle in the incubator for a short time.
- Remove the spent medium from the culture vessel (flask, petri dish, etc.).
- Add a small amount of dissociation solution to the vessel without disturbing the monolayer. This is usually best performed by slowly adding the solution to the side of the flask opposite the monolayer or to the side of a petri plate. Gently swirl the vessel to cover the monolayer with the solution. Discard the solution from the vessel.
- Repeat Step #3. Leave a small amount of Cellstripper® solution in the vessel, enough just to cover the monolayer.
- Incubate the cells at 37°C, observing microscopically, until the cells begin to round up. Tapping the side of the vessel may facilitate removal of difficult cell lines.
- After detachment, disperse the cells into suspension by pipetting repeatedly. Continue with the normal procedure.
