Wide-pore bonded phase packings used in these analytical columns are based on spherical 5mm silica gel with a mean pore diameter of 300Å. To this base are bonded a variety of functionalities to accomplish separations of a wide range of biomolecules. These packings are chemically stable over the pH range 2 to 8.5 and to such cleaning and antimicrobial agents as sodium azide, dilute trifluoroacetic acid and acetic acid. They are also unharmed by organic solvents such as methanol and acetonitrile and by concentrated aqueous solutions of salts and urea.
Wide-pore packings are available in the following chemistries:
- Octadecyl (C18), the most retentive reversed phase packing for effective separations of peptides and hydrophilic proteins.
- Octyl (C8), for moderately hydrophobic proteins and other moderately nonpolar compounds. Use when octadecyl packing is too retentive or when faster analyses are needed.
- Butyl (C4) for quantitative recoveries of proteins over a wide range of hydrophobicities. Provides high resolution and rapid analysis.
- HI-Propyl (CH2CH2CH3), hydrophobic interaction (HI) medium for the fast resolution of biopolymers on the basis of differences in weak hydrophobic interactions in an aqueous mobile phase for retention of full biological activity. Quantitative protein recoveries for most proteins.
The standard 4.6mm x 25cm column configuration provides the highest resolution and a typical operating back pressure of 1000 to 1500 psi at a flow rate of 1.0mL/min using aqueous mobile phase.
