General description: | Organisms known to grow in single strength and concentrated juices are lactic acid and acetic acid bacteria and yeast. Lactobacilli, Leuconostoc and yeast have all been identified as spoilage organisms by numerous authors. Orange serum at pH 5.4 to 5.6 has been reported to yield maximum counts of all types of spoilage organisms in mixed cultures and in single culture comparison tests. The low pH of the test solution usually prohibits the development of other microorganisms which cannot survive low pH conditions. Therefore developing colonies are presumed to be problematic organisms. Formulation. Per liter of water, adjusted to pH 5.6 at 25oC • Orange serum 100ml • Yeast extract 3.0g • Tryptone 10.0g • Dextrose 4.0g • Dipotassium phosphate 2.5g |
