Preparation Note: | Activator: NaCI (10 mM) activates the pancreatic enzyme. Stabilizers: Both, the bacterial and the pancreatic enzymes are stabilized by Ca2+ (bound Ca2+ is required for activity). Working concentration: 2.5 to 5 mg/ml Working solution: Phosphate buffer (100 mmol/l; pH 7.0). |

Unit Definition: | Incubation procedure: M13mp9ss, M13 primer (17mer) and 1 μCi (α32P) dCTP are incubated with suitable dilutions of Taq DNA Polymerase in 50 μl incubation buffer at +65 °C for 60 minutes. The amount of incorporated dNTPs is determined by trichloroacetic acid precipitation. |
