Application: | Streptavidin-POD has been used for: • ELISA • In situ hybridization • Western blot It is also used for immunohistochemistry. |
General description: | Streptavidin is covalently coupled to horseradish peroxidase. The conjugate is used for detection of biotin-labelled substances. |
Other Notes: | For life science research only. Not for use in diagnostic procedures. |
Physical form: | Lyophilizate, stabilized |
Preparation Note: | Working concentration: 5 - 500 mU/ml (i.e., stock solution can be diluted). Working concentration of conjugate depends on application and substrate. The following concentrations should be taken as a guideline: • ELISA: 5 to 50 mU/ml • Immunohistocytochemistr • Western blot: 100 to 250 mU/ml |
Reconstitution: | Add 1 ml double-distilled water to a final concentration of 500 U/ml. The reconstituted stock solution is stable at 2 to 8 °C for 6 months. Do not freeze reconstituted antibody! Do not add sodium azide! A further dilution of reconstituted stock solution in double-distilled water is stable for approx. one week at 2 to 8 °C. |
Unit Definition: | One unit Streptavidin-POD, corresponding to one unit peroxidase, will oxidize 1 μmol of guaiacol tetramer in one minute at +25 °C and pH 7.0. Unit Conversion: 1 U (+25 °C; guaiacol as substrate; pH7.0) corresponds to 1.24 U (+25 °C; 1.7 mM ABTS as substrate; pH= 6.0) or to 4.0 U (+25 °C; 8.7 mM ABTS as substrate; pH = 5.0). |
