Application: | Semimicro method for the determination of nitrite and/or nitrate to investigate water, waste water, environmental samples, plant material, foodstuffs, drugs, cosmetics, and biological samples. Nitrite/Nitrate, Colorimetric Test was used in a study done to estimate the in vivo effects of myrtle oil (Myrtii oleum) on the antioxidant enzymes such as catalase and superoxide dismutase in alloxan and normoglycaemic diabetic rabbits. |

Preparation Note: | Working solution: Preparation of the solutions • Use the contents of bottle #1 undiluted, bring to 15 to 25 °C before use. • Place one Coenzyme-tablet in a beaker and dissolve in 3 ml of the solution from bottle 1. Remove the Coenzyme-tablet using the tweezers provided. The resulting solution is reaction mixture 2 and is sufficient for 12 nitrate determinations, bring to 15 to 25 °C before use. • Dissolve the contents of one bottle #3 in 0.7 ml of double-dist. water to give solution 3. • Use the contents of one bottle #4 undiluted. • Use the contents of one bottle #5 undiluted. Storage conditions (working solution): • Prepare Reaction Mixture 2 immediately before use. • Solution of Nitrate reductase is stable at 2 to 8 °C for 2 weeks. • Solution of Color reagent I is stable at 2 to 8 °C for 3 months. • Solution of Color reagent II is stable at 2 to 8 °C for 3 months. |
