X-tremeGENE 9 DNA Transfection Reagent
An effective balance of high efficiency and low cytotoxicity. CHO-K1 cells were transfected with a GFP expression plasmid with two different transfection reagents using the reagent/DNA ratio recommended by each of the suppliers. Panels A and C show the fluorescent signals after transfection. Panels B and D are the corresponding bright field photomicrographs. After transfection with LTX P, fluorescent signals are visible, but there are fewer viable cells compared to transfection using X-tremeGENE 9 Transfection Reagent. Cell imaging and analysis was performed using the Roche Cellavista System.
Cytotoxicity comparison with competitor reagents. Reagent cytotoxicity was measured using the Roche LDH Cytotoxicity Detection Kit (LDH), after transfecting HeLa cells with X-tremeGENE 9 DNA Transfection Reagent and two different competitor reagents. The X-tremeGENE 9 DNA Transfection Reagent showed the lowest cytotoxic effect.
Transfection efficiency comparison with reference transfection reagent. Four widely used cell lines were transfected in serum-containing medium using X-tremeGENE 9 Transfection Reagent. Transfection efficiencies were measured as a ratio of GFP-transfected cells versus whole cell number using the Roche Cellavista System, and normalized to the reference transfection reagent set at 1. Error bars show the standard deviation of the mean of triplicates.
High cell survival and reporter gene expression HeLa cells were transfected using X-tremeGENE 9 Transfection Reagent and two different competitor reagents. Reagent cytotoxicity was measured using the Roche LDH Cytotoxicity Detection Kit (LDH). X-tremeGENE 9 DNA Transfection Reagent showed the highest transfection efficiency and the lowest levels of cytotoxicity.

During quality testing, cells are transfected with a reporter gene vector DNA using X-tremeGENE 9 DNA Transfection Reagent (ratio 3:1 μl/μg DNA). Reporter gene activity is monitored via chemiluminescent detection. Using a standard curve, the total amount of recombinant protein is determined per well in order to meet specification.
