X-tremeGENE HP DNA Transfection Reagent
An effective balance of high efficiency and low cytotoxicity. CHO-K1 cells were transfected with a GFP expression plasmid, with two different transfection reagents using reagent/DNA ratios recommended by each of the suppliers. Panels A and C show the fluorescent signals after transfection. Panels B and D are the corresponding bright field photomicrographs. After transfection with LTX P, fluorescent signals are visible, but there are fewer viable cells compared to transfection with X-tremeGENE HP Transfection Reagent. Cell imaging and analysis was performed using the Roche Cellavista System.
X-tremeGENE HP DNA Transfection Reagent outperforms the competitor reagent. Four hard-to-transfect cell lines were transfected in serum-containing medium using X-tremeGENE HP Reagent. Transfection efficiency was measured as a ratio of GFP-transfected cells versus whole cell number using the Cellavista System and normalized to the reference reagent set at 1. Error bars show the standard deviation of the mean of triplicates.
Performance comparison with competitor reagents. Comparison using a luciferase encoding plasmid in HeLa cells with transfection reagent to DNA ratios as recommended by the respective suppliers is shown. Error bars show the standard deviation of the mean of triplicates.
Cell survival comparison with competitor reagents. HeLa cells were transfected with X-tremeGENE HP Transfection Reagent and two different competitor reagents. Reagent cytotoxicity was measured using the Roche LDH Cytotoxicity Detection Kit (LDH). The applied ratios of microliter reagent to microgram DNA are indicated. X-tremeGENE HP DNA Transfection Reagent showed the lowest levels of cytotoxicity.

During quality testing, cells are transfected with a reporter gene vector DNA using X-tremeGENE HP DNA Transfection Reagent (ratio 3:1 ¼l/¼g DNA). Reporter gene activity is monitored via chemiluminescent detection. Using a standard curve, the total amount of recombinant protein is determined per well in order to meet specification.
