General description: | Legionella in Water - Water Pollution (WP) is produced in accordance with ISO/IEC 17043:2010. The PT portfolio allows our test components to offer significant performance data to monitor and prove competency to customers, accreditation bodies or both. Preparation Note: | Description• This PT sample is designed for the quantitative analysis of Legionella in water. The PT sample is provided in duplicate as a stable dehydrated disc requiring hydration. The additional sample is provided for mishaps in the lab. This sample requires 100 ml of buffer/ saline solution for hydration fluid (see note below). The dehydrated disc sample is stable for a time exceeding the study schedule. General Information• Due to the sensitivity of microorganisms and short holding times after hydration, please read entire instructions prior to preparation. This sample is a single disc and the possible concentration range is quite broad, it may be necessary to analyze the sample in a dilution series using general phosphate buffer solution. The hydration temperature of 35-37°C is rigorous. Low temperature will not assure complete hydration. Higher temperature will kill the microorganisms. Sample Preparation• Allow the capped sample vial to equilibrate to room temperature. Open the sample vial and aseptically transfer the disc to the hydrating fluid. • The sample is the colored disc within the cryotube. It will either be on top of cotton with opaque desiccant beads below the cotton or free moving in a cryotube that contains a desiccant tablet within the cap. After 15 min inspect to assure dissolution of the disc. It should dissolve completely. This represents the sample for analysis. Dilutions are recommended to achieve a proper count, but base all counts on the whole volume sample. NOTE: The Vitroid™ is the colored disk within the cryotube. It will either be on top of cotton with opaque desiccant beads below the cotton or free moving in a cryotube that contains a desiccant tablet within the cap. The opaque desiccant beads and desiccant tablet are to keep the container dry and are not biologicals. • Shake well and place the sample in your incubator for 15 minutes. The hydration temperature of 35 - 37 °C is rigorous. • Low temperature will not assure complete hydration. • Higher temperatures will kill the microorganisms. After 15 minutes, remove the hydrated sample from the incubator and shake well. Inspect to assure dissolution of the Vitroid™. The disk will dissolve completely. • This represents the sample for analysis. Dilutions are recommended to achieve a proper count but base all counts on the whole volume sample. Note: We are no longer supplying Phosphate buffer for Micro samples. Please proceed to use your laboratory′s standard buffer or saline solution. If your laboratory does not have buffer in-house, we recommend using P3813, Phosphate Powder which can be purchased at SigmaAldrich.com. |
