General description: | This Whole Effluent Toxicity (WET) proficiency testing sample is produced in accordance with ISO/IEC 17043:2010. This product is produced according to TNI requirements. However, please note, this sample may include non-TNI analytes as well. In Acute WET tests, Fathead minnow is employed as a test organism to determine its mortality upon exposure to the effluent for a short duration (48 h). This helps arrive at the lethally toxic concentrations of the effluent. Preparation Note: | Description • The effluent is provided as a concentrate for dilution in moderately hard synthetic freshwater (MHSF). • This concentrate is an EPA-specified toxicant in reagent-grade DI water. • Enough material has been provided to run the method at least twice. If you require more toxicant, please contact ptservice@milliporesigma. • Do not deviate from the supplied instructions or EPA Method 2000.0. Contact ptservice@milliporesigma. Preparation Instructions • To prepare 1 L of effluent, add 900 mL of MHSF to a volumetric flask. Preparation instructions for MHSF can be found in the EPA document EPA-821-R-02-012, page 32. • Transfer the entire contents of one ampule to the flask. Rinse the ampule with MHSF and transfer to the volumetric flask. Bring flask to 1 L volume with MHSF. • The effluent prepared according to these instructions represents the sample ready for testing, 100% effluent. This 100 % effluent is split into two 0.5 L aliquots. The first aliquot is your 100% effluent for testing. The second aliquot is diluted with 0.5 L MHSF and mixed. This becomes your 50% effluent sample. Continue diluting half of each sample with the same volume of MHSF to make your 25%, 12.5% and 6.25% effluent dilutions, which represent the five test dilutions. Required Test Conditions • Test Type: Static non-renewal • Test duration: 48 h,Temperature: 25 ºC ±1 ºC. The test must not deviate by more than 3 ºC during the test. • Renewal of Test solutions: non-renewal • Dilution water: Moderately hard synthetic water. Test Conditions • Light quality: Ambient laboratory illumination (recommended) • Light intensity: 10-20 μE/m2/s (50-100 ft-c) (ambient laboratory levels) (recommended) • Photoperiod: 16 h light, 8 h darkness (recommended) • Test chamber size: 250 mL (recommended minimum) • Test solution volume: 200 mL (recommended minimum) • Age of test organisms: 1-14 days; less than or equal to 24-h range in age (required), • No. organisms per test chamber: 10 (required minimum) • No. replicate chambers per concentration: 2 (required minimum) • No. of organisms per concentration: 20 (required minimum) • Feeding regime: Artemia nauplii are made available while holding prior to the test • Test chamber cleaning: Cleaning not required • Test solution aeration: None, unless DO concentration falls below 4.0 mg/L; rate should not exceed 100 bubbles/min. • Test concentrations: 5 and a control (required minimum) • Dilution series: greater than or equal to 0.5 dilutions series (recommended) • Test acceptability criterion: 90% or greater survival in controls • Sample holding requirements: Composite sample within 36 h of effluent preparation (required) |
