Enhanced Validation-By Independent Antibodies Immunofluorescence. HepG2 cells were fixed and permeabilized with 4% paraformaldehyde followed by 0.5% Triton X-100. Fixed cells were stained with 1:500 Anti-l-Afadin antibody produced in Rabbit (Cat. No. A0349) (A). The antibody developed using 1:50 Goat Anti-Rabbit IgG (whole molecule)-FITC conjugate antibody, and 1:500 Anti-l/s-Afadin antibody produced in Rabbit (Cat. No. A0224) (B). The antibody was developed using Goat Anti-Rabbit IgG, FITC Conjugate antibody. Results Two Anti-l/s-Afadin antibodies, A0349 (A) and A0224 (B) target different regions of l/s-Afadin show similar staining profiles between the two antibodies, demonstrating Independent Antibody Verification.
Immunohistochemistry Frozen sections, acetone fixed Mouse liver were stained with 1:1,000 Anti-l-Afadin antibody produced in Rabbit (Cat. No. A0349), followed by 1:50 Anti-Rabbit IgG (whole molecule)-FITC antibody produced in Goat (Cat. No. F9887).
Western Blotting Whole extract of Rat brain S1 were separated on SDS-PAGE and probed with Anti-l-Afadin antibody produced in Rabbit (Cat. No. A0349). The antibody was developed using 1:10,000 Anti-Rabbit IgG (whole molecule)-Peroxidase antibody produced in Goat (Cat. No. A0545). Biochem/physiol Actions: AF-6/Afadin regulates the formation of tight and adherens junctions in the cell and its absence in mice leads to embryonic defects and lethality . Afadin is implicated in primary breast cancer and pancreatic cancer. It downregulates the process of migration and metastasis in cancer cells.

General description: Afadin is a multidomain protein that binds F-actin and E-catenin proteins at cell-cell adheren junctions. I-afadin is a ubiquitously expressed splice variant of afadin that contains PDZ and F-actin binding domains at the C-terminal end. I-afadin is homologous to the human AF-6 protein .
Immunogen: Synthetic peptide sequence corresponding to amino acid residues 1814-1829 of rat afadin with N-terminal cysteine conjugated to maleimide-activated KLH.
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 1% BSA and 15 mM sodium azide
Specificity: Anti-l-Afadin antibody is specific for I-afadin (approximately 200 kDa). It also binds I-afadin in rat, mouse, dog and human.
