Immunofluorescence HeLa cells were fixed and permeabilized with cold methanol followed by cold acetone. Fixed cells were stained with 1:200 Rabbit Anti-ATG4B (Cat. No. A2981). The antibody was developed using Goat Anti-Rabbit IgG, ATTO-488 conjugate. Cells were counterstained with DAPI (blue) to stain nuclei.
Immunoblotting Whole mouse 3T3 cell extract was separated on SDS-PAGE and blotted with Rabbit Anti-ATG4B (Cat. No. A2981). The antibody was developed with Goat Anti-Rabbit IgG, Peroxidase conjugate (Cat. No. A0545) and a chemiluminescent substrate. Lanes 1: Antibody 1:250 2: Antibody 1:500 3: Antibody 1:500 + ATG4B immunizing peptide (human 6-22)

Biochem/physiol Actions: ATG4B (autophagy-related 4B cysteine peptidase) participates in the modulation of autophagy in mammalian cells. It is also involved in light chain 3 (LC3) lipidation. ATG4B is responsible for the processing of the mammalian Atg8 homologues: Golgi-associated ATPase enhancer of 16 kDa (GATE16), gamma-aminobutyric-acid-type-A-receptor-associated protein (GABARAP), and LC3. It removes the carboxyl-terminal region of proLC3 immediately after its synthesis, generating the soluble LC3-I and exposing a carboxyl-terminal Gly.

General description: ATG4B (autophagy-related 4B cysteine peptidase), a mammalian orthologue of yeast Atg4, is a cysteine protease. It is widely expressed in human and rat tissues, being most abundant in the brain. This gene is mapped to human chromosome 2q37.
General description: ATG4B is a member of the C-54 family of cysteine proteases that is involved in cell degradation through autophagy.
Immunogen: synthetic peptide corresponding to amino acids 6-22 of human ATG4B, conjugated to KLH via a C-terminal cysteine residue. The corresponding sequence is identical in mouse.
