Application: Achromopeptidase from bacteria may be used:
• in the homogenization step for genomic DNA extraction from microbial biofilm
• in the lysis of gram-positive strains(
• for DNA extraction from Nocardia strains

Application: Achromopeptidase is useful for lysis of Gram-positive bacteria that are resistant to lysozyme. Achromopeptidase, from Sigma, has been used for the isolation and molecular characterization of skin and soft-tissue bacterial infections. Product A3422 is from bacteria and is a partially purified powder containing lactose.
Biochem/physiol Actions: Achromopeptidase is a bacteriolytic enzyme and belongs to serine protease family. It is effective for the lysis of gram positive bacteria compared to lysozyme. Achromopeptidase activity is inhibited in the presence of heavy metals. Achromopeptidase based inhibition assay may be useful for screening the presence of heavy metals.
Biochem/physiol Actions: Achromopeptidase is a lysyl endopeptidase that has a molecular weight of approximately 27 kDa. It works best between pH 8.5 -9. Approximately 500-1,500 μn/ml achromopetidase can be used to lyse cells at a density of OD600 = 0.6 over 2 hours at 37 °C.

Unit Definition: One unit will produce a change in A600 of 0.001 per minute per mL at pH 8.0 at 37 °C using a suspension of Micrococcus lysodeikticus as substrate (1 cm light path).
