Enhanced Validation-By Independent Antibodies Formalin fixed, paraffin-embedded, methacarn-treated sections of Human Alzheimer’s disease (AD) brain were stained with 1:100 Anti-β-Amyloid Protein (1-40) antibody produced in Rabbit (Cat. No. A8326) (A) and developed using 15 μg/mL Anti-Rabbit IgG (whole molecule)-Biotin antibody produced in Goat (Cat. No. B8895) and by 20 μg/mL ExtrAvidin®-Peroxidase (Cat. No. E2886), and 2.5 μg/mL Anti-β-Amyloid antibody, Mouse monoclonal, Clone: BAM-10 (Cat. No. A3981) (B). The antibody was developed using using 15 μg/mL Anti-Mouse IgG (whole molecule)-Biotin antibody produced in Goat (Cat. No. B7151) and by 20 μg/mL ExtrAvidin®-Peroxidase (Cat. No. E2886). Results Two Anti-β-Amyloid antibodies,A8326 (A) and A3981 (B) target different regions of β-Amyloid show similar staining profiles between the two antibodies, demonstrating Independent Antibody Verification.
Immunohistochemistry Formalin fixed, paraffin-embedded, methacarn-treated sections of Human Alzheimer’s disease (AD) brain were stained with 1:100 Anti-β-Amyloid Protein (1-40) antibody produced in Rabbit (Cat. No. A8326), followed by 15 μg/mL Anti-Rabbit IgG (whole molecule)-Biotin antibody produced in Goat (Cat. No. B8895) and by 20 μg/mL ExtrAvidin®-Peroxidase (Cat. No. E2886).
Immunoblotting Beta-Amyloid (Sig. A5813) was separated on SDS-PAGE and probed with AB to Beta-Amyloid (1-40) (RB) (Prepack) (Cat. No. A8326). The antibody was developed with Alk Phos APA Rabbit Igc (Cat. No. A9919) and NBT/BCIP substrate. Antibody dilution 1/1,000.

Biochem/physiol Actions: The β-amyloid precursor protein (APP) is cleaved sequentially by the proteolytic enzymes β-secretase (BACE1) and γ-secretase to produce β-amyloid (Aβ) peptides with the Aβ1-42 and the Aβ1-40 forms being the most prevalent. Secreted Aβ peptides are degraded either via a re-uptake mechanism followed by endosomal degradation, or by an extracellular insulin degrading enzyme. Extracellular accumulation of Aβ leads to the formation of aggregates, fibrils and eventually amyloid deposits called neuritic plaques, which is the hallmark of Alzheimer′s disease (AD).
Rabbit Anti-β-Amyloid Protein (1-40) antibody does not stain control sections of normal brain tissues.

General description: Amyloid precursor proteins (APPs) are members of a large family of 70 kDa transmembrane glycoproteins that are found in a wide range of tissues. APP is expressed in the brain. It is located on human chromosome 21. APPs have three main isoforms, namely, APP695, APP751 and APP770, that are derived from alternative splicing events in cells.
Immunogen: synthetic β-amyloid (1-40) conjugated to BSA.
Physical form: Rabbit Anti-β-Amyloid (1-40) is supplied as a liquid containing 0.1% sodium azide as preservative.
