Application: Anti-GCLC antibody produced in rabbit is suitable for western blotting at a concentration of 1μg/mL.
Biochem/physiol Actions: GCLC gene encodes the heavy catalytic subunit of the enzyme glutamate-cysteine ligase, which is the rate limiting enzyme of glutathione synthesis. Mutation at this locus is associated with hemolytic anemia due to deficiency of γ-glutamylcysteine synthetase. Polymorphism in GCLC subunit is found to be associated with sulfamethoxazole-induced hypersensitivity in HIV/AIDS patients in a study. A GAG-trinucleotide repeat polymorphism in the 5′-untranslated region of the gene is shown to lower the levles of GCL activity and GSH (glutathione), a major intracellular antioxidant. Therefore, it is associated with increased risk for lung and aerodigestive tract cancers.
