Immunofluorescence
Proliferating cells in the eye of E4 chick embryo were stained with Monoclonal Anti-BrdU, clone no. BU-33 (Cat. No. B2531) and developed using Rabbit Anti-Mouse IgG (whole molecule)-FITC.
From R.S. Goldstein, Dept. of Life Science, Bar-Ilan University, Ramat Gan, Israel.
Immunohistochemistry
Triple sections from E4 chick embryo: Nervous system (pink) is stained with Monoclonal Anti-HNK-1/N-CAM, clone no. VC1.1 (Cat. No. C0678) and Goat Anti-Mouse IgG-Peroxidase (Cat. No. A3673). Nuclei that incorporated BrdU are stained with Monoclonal Anti-BrdU, clone no. BU-33 (Cat. No. B2531) and Goat Anti-Mouse IgM-Alkaline Phosphatae (Cat. No. A7784). BrdU-negative nuclei were stained with Harris′ hematoxylin (Cat. No. H5522). From Dr. R.S. Goldstein, Dept. of Life Sciences, Bar-Ilan university, Ramat Gan, Israel.
Hetrokaryons between proliferating (BrdU pre-labeled) and quiescent (unlabeled) endothelial cells were treated with TNF-α to induce programmed cell death (apoptosis). BrdU incorporation into DNA was detected with Monoclonal Anti-BrdU, (clone no. BU-33, Cat. No. B2531) followed by goat Anti-Mouse IgG-β-galactosidase. After staining with acridine orange (Cat. No. A6014) cells were visualized by phase contrast (left) and fluorescent (right) microscopy. Tailed arrows indicate intact (solid) or apoptotic (dotted) BrdU positive nuclei, arrow heads point to normal (solid) and apoptotic (notched) BrdU negative nuclei. From V. Polunovsky, Univ. of Minnesota, Minneapolis, MN.
Immunohistochemistry
Neural tube (spinal cord) of E4 chick embryo stained with Monoclonal Anti-BrdU (clone no. BU-33, Cat. No. B2531) and hematoxylin.
From Dr. R.S. Goldstein, Dept. of Life Sciences, Bar-Ilan University, Ramat Gan, Israel.
Staining of BrdU from 4th embryonic day (E4) chicks using Monoclonal Anti-BrdU (clone no. BU-33, Cat. No. B2531). Spinal cord. From Dr. R. Goldstein, Bar-Ilan University, Ramat Gan, Israel.
Staining of BrdU from 4th embryonic day (E4) chicks using Monoclonal Anti-BrdU (clone no. BU-33, Cat. No. B2531). Eye. From Dr. R. Goldstein, Bar-Ilan University, Ramat Gan, Israel.

technique(s) immunocytochemistry: suitable
immunohistochemistry (formalin-fixed, paraffin-embedded sections): 1:1000 using sections of intestine from mouse or rat treated in vivo with BrdU
Application: Monoclonal Anti-BrdU antibody has been used in immunostaining, BrdU incorporation into DNA, immunohistochemistry and uterine labeling index. It has also been used to investigate cell proliferation.
Biochem/physiol Actions: 5′-Bromo-2-deoxyuridine (BrdU) staining is predominantly used to observe the multiplication of tumor cells and other tissues in vivo.
Biochem/physiol Actions: Reacts specifically with BrdU (bromodeoxyuridine) incorporated into DNA or coupled to a protein carrier. Cross-reaction with IdU (iododeoxyuridine) is observed.

General description: 5′-Bromo-2-deoxyuridine (BrdU) is a carcinogen that may be absorbed by skin or inhalation. BrdU solutions are sensitive to light. Monoclonal anti-BrdU (immunohistology grade) (mouse IgG1 isotype) is derived from the BU-33 hybridoma produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with bromodeoxyuridine conjugated to KLH.
Immunogen: bromodeoxyuridine conjugated to KLH.
Other Notes: This product can be found as purified product that was produced using cell culture hybridoma product.
B8434 Anti-BrdU antibody, Mouse monoclonal
clone BU-33, purified from hybridoma cell culture
Preparation Note: For continuous use, store at 0-5EC. For extended storage, the solution may be frozen in working aliquots. Repeated freezing and thawing is not recommended. Storage in "frost-free" freezers is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use.
