Immunoprecipitation Lanes 1, 2, 3. 5, 2, 5 and 1.0 μg of Rabbit Anti-53BP1 (C-ter), Cat. No. B4436 was used to immunoprecipitate human 53BP1 from 0.25 mg of 293T cell lysates; the immunoprecipitation was detected with antibody. rabbit anti 53BP1 (N-ter), Cat. No. B4561, and developed with Goat anti-Rabbit IgG, Peroxidase conjugate followed by a chemiluminescent substrate. 4. Negative control — without cell lysate 5. Negative control — without IP antibody 6. Positive control — Extract of 293-T cells was separated on SDS-PAGE and blotted with Rabbit Anti-53BP1 (N-ter) (Cat. No. 4561)
Western Blotting U2OS total cell extracts were separated on SDS-PAGE and probed with Anti-53BP1 (C-terminal) antibody produced in Rabbit (Cat. No. B4436) followed by 1:30,000 Goat Anti-Rabbit IgG-Peroxidase (Cat. No. A0545). Lanes 1. 4 μg/mL antibody 2. Negative Control

Biochem/physiol Actions: Tumor protein p53 binding protein (T153BP1) has a close link to ataxia-telangiectasia mutated (ATM)-regulated events. 53BP1 becomes phosphorylated by ATM in a DNA damage-dependent manner suggesting that it participates in propagating the ATM signaling to its downstream effectors. T153BP1 also plays a key role in DNA repair by mediating nonhomologous end-joining (NHEJ) near double stranded breaks (DSBs). Fusion of T153BP1 with platelet-derived growth factor receptor beta (PDGFRβ) is implicated in a type of myeloproliferative disorder accompanied with eosinophilia.

General description: Tumor protein p53 binding protein (T153BP1) is mapped to human chromosome 15q15.3. It has methylated histone-binding module and tandem tudor domains.
Immunogen: synthetic peptide corresponding to amino acids 1955-1972 of human 53BP1, conjugated to KLH via an N-terminal added cysteine residue. This sequence is conserved in mouse, and differs from rat by one amino acid
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

New Tariffs & Effects on Pricing

Tariff note: As we navigate the current environment of tariff surcharges and mid-year manufacturer price increases, we want to keep you informed. While we have avoided making specific decisions thus far, we anticipate that we will be forced to implement a cost adjustment policy to offset the fees we have already been absorbing.
