Immunofluorescence
Coronal section of the developing cerebral cortex of an embryonic day 16 rat stained with Monoclonal Anti-Chondroitin Sulfate, clone no. CS-56 (Cat. No. C8035) followed by fluorescein labeled Goat Anti-Mouse Ig. The chondroitin sulfate proteoglycan (CSPG) is most concentrated in the pia matter and the sub-plate (bright green layers). There is moderate staining in the intermediate zone, and there is very little CSPG in the cortical plate and ventricular zone. The cortex was stained with bis-benzimide (Cat. No. B2883) that labels the cell nucleus and fluoresces blue under ultraviolet illumination.

Immunofluorescence BMGE cells were fixed and permeabilized with 4% paraformaldehyde followed by 0.5% Triton X-100. Fixed cells were stained with 1:200 Monoclonal Anti-Chondroitin Sulfate, Clone: CS-56 (Cat. No. C8035). The antibody was developed using Goat Anti-Mouse IgG, Cy3™ conjugate. Cells were counterstained with DAPI (blue) to stain nuclei.

Monoclonal Anti-Chondroitin Sulfate can also be used to study chondroitin sulfate proteoglycan (CSPG) distribution and its relationships to specific cell-substrate contacts. It is also suitable for microarray and indirect immunofluorescence at the dilution of 1:200 by using bovine mammary gland epithelial (BMGE) cells. |
| Biochem/physiol Actions: | Chondroitin sulfate is involved in tumor invasion and also plays a vital role in stabilizing cell-substratum interactions during early events of melanoma cell spreading on endothelial basement membranes. In Chondroitin sulfate proteoglycan (CSPG), proteoglycan part is involved in cell proliferation and migration which stimulates endothelial cells motility during microvascular morphogenesis. Chondroitin sulfate is used in osteoarthritis treatment. Upregulation of chondroitin sulfate proteoglycans is observed after neuronal injury. It possess the ability to inhibit axonal regeneration. |
| Biochem/physiol Actions: | The antibody has been reported to be specific for the glycosaminoglycan (GAG) portion of native chondroitin sulfate proteoglycan (CSPG). The antibody reacts specifically with chondroitin sulfate types A and C but not with type B (dermatan sulfate), and may be used for localization of chondroitin sulfate in tissues and cultured fibroblasts. |
