Principle of assay N-Acetyl-β-D-Glucosaminidase=NAG 4-Nitrophenyl-N-acetyl-β-D-glucosaminide= NP-GlcNAc The NAG enzyme hydrolyzes the substrate (NP-GlcNAc) at acidic environment (pH ~4.7), during incubation at 37°C. The enzymatic hydrolysis of the substrate liberates p-Nitrophenylate ion. The p-Nitrophenylate ion becomes yellow at basic environment (pH 10-11), and the reaction product is measured calorimetrically at 405nm.

| Analysis Note: | The kit assay is based on the hydrolysis of NAG substrate (NP-GLcNAc) by the enzyme. The enzymatic hydrolysis of the substrate liberates p-nitrophenylate ion. The reaction product is detected colorimetrically at 405 nm. |
