Principle of Diaminofluoresceins as Indicators of Nitric Oxide or Nitric Oxide Synthase Activity. When the DAF2-DA dye penetrates cells, it is hydrolyzed by intracellular esterase activity to produce the parent DAF2 compound, which reacts rapidly with NO produced by NOS to form a fluorescent triazolofluorescein (DAF-2T). The fluorescent product can be quantitated by fluorometry using an excitation filter of 492 nm and an emission filter of 515 nm. It can also be visualized with standard fluorescein filter set by microscopy.
Nitric Oxide Visualization in Living Cells Using DAF-2DA (D225). LSP/γ-INF induced RAW 264.7 cells were stained with DAF-2DA and visualized by fluorescence microscopy.

General description: 4,5-Diaminofluorescein diacetate (DAF-2 DA) solution is a membrane-permeable fluorescent probe, which measures nitric oxide (NO) in living cells. It is used to study the synthesis of NO in bovine aortic endothelial cells (BAECs). DAF-2 DA imaging shows that ciliated epithelial cells produces high amount of NO than non-epithelial inflammatory cells.
