Immunohistochemistry
Lung carcinoma labeled with Monoclonal Anti-Epidermal Growth Factor Receptor (Cat. No. E2760).
From R. Robinson, Univ. of Iowa, College of Med., Dept. of Pathology, Iowa City, IA.
Immunofluorescence
Photomicrograph of a mixed cell culture of an amelanotic melanoma cell line (Hs695T) and an epidermoid carcinoma cell line (A431). Cells were grown on coverslips, fixed and stained with Monoclonal Anti-EGF Receptor, clone no. 29.1 (Cat. No. E2760) – green fluorescence; and Hoechst No. 33258 (Cat. No. B 2883) – blue nuclear stain. Coverslips were photographed on a NIKON Microphot-FXA.
From G. MacKenzie, Immunogen, Inc., Cambridge, MA.
Western Blotting Whole extract of A431 cells were separated on SDS-PAGE and probed with Monoclonal Anti-Epidermal Growth Factor Receptor antibody produced in Mouse, Clone: 29.1 (Cat. No. P2859). The antibody was developed using 1:10,000 Anti-Mouse IgG (Fab specific)-Peroxidase antibody produced in Goat (Cat. No. A9917). Lanes 1. 1:6,000 antibody 2. 1:3,000 antibody 3. Negative Control
ELISA Monoclonal Anti-Epidermal Growth Factor Receptor antibody produced in Mouse, Clone: 29.1 (Cat. No. E2760), followed by 1:1,000 Anti-Mouse IgG (Fab specific)-Peroxidase antibody produced in Goat (Cat. No. A9917), was serially diluted to detect Epidermal Growth Factor Receptor by indirect ELISA.

Biochem/physiol Actions: Localizes blood type A-related determinants on various glycoproteins and glycolipids in addition to specific EGF receptor determinants (only observed in human tissue). Binds an external carbohydrate on the receptor and does not block EGF binding.
Biochem/physiol Actions: The members of epidermal growth factor receptor (EGF R) or the ErbB receptor family have been identified as useful biomarkers and targets for cancer therapy. The EGFR family includes four receptor tyrosine kinases, EGF R (ErbB1), ErbB2 (Neu), ErbB3, and ErbB4. EGF R binds EGF and induces tyrosine phosphorylation leading to proliferation of cells. EGF R is present on many cell types of epithelial and mesenchymal lineages. EGF R is capable of binding transforming growth factor-α and heparin-binding EGF in addition to EGF. There are numerous effector molecule activated by EGF R that result in a variety of biological processes such as morphogenesis, cell motility, apoptosis, differentiation and organ repair and maintenance. Deregulation of EGF R signaling is implicated in progression of a wide variety of tumors, invasion and metastasis.

General description: Monoclonal Anti-Epidermal Growth Factor (EGF) Receptor (mouse IgG1 isotype) is derived from the hybridoma produced by the fusion of mouse myeloma cells and splenocytes from an immunized mouse. The receptor for epidermal growth factor is an integral cell membrane protein of 170 kDa, which spans the membranes of a wide range of normal and malignant epithelial cells. A close similarity has been found between the sequence of the avian erythroblastic leukemia viral oncogene homolog (v-erb-B) and the cytoplasmic and transmembrane part of the EGF-R (truncated EGF-R).
Immunogen: human carcinoma cell line A431, which expresses high levels of EGF receptors.
