Immunoblotting Analysis of lysates from several cell lines, using eIF4ENIF1 antibody: The extracts were separated on SDA-PAGE and blotted with Rabbit Anti-eIF4ENIF1, Affinity isolated antibody (Cat. No. E6656), at a concentration of 2 μg/mL. The antibodies were developed with Goat Anti-Rabbit IgG, Peroxidase conjugate (Cat. No. A0545) and a chemiluminiscence substrate. Biochem/physiol Actions: Anti-eIF4ENIF1 specifically recognizes human eIF4ENIF1.
Biochem/physiol Actions: eIF4ENIF1 (also known as 4E-T or 4E-transporter) colocalizes with other mRNA binding/processing proteins to processing (P)-bodies. The interaction of eIF4ENIF1 with eIF4E in the cytoplasm represses translation, which is believed to be a prerequisite for targeting of mRNAs to P-bodies. eIF4ENIF1 is likely to inhibit translation by competing with eIF4G for binding to eIF4E and preventing the formation of the eIF4F complex. Together with 4E-BPs, it participates in cell response to prolonged hypoxia, sequestering eIF4E, and affecting gene expression. A nonsense mutation in the eIF4ENIF1 gene is implicated in the Primary Ovarian Insufficiency.
Biochem/physiol Actions: Eukaryotic translation initiation factor 4E transporter (4E-T) is a protein encoded by the EIF4ENIF1 gene in humans. It is a nucleocytoplasmic shuttling protein that mediates the import of eIF4E into the nucleus. 4E-transporter (4E-T) is one of several proteins that binds the mRNA 5′cap-binding protein, eukaryotic initiation factor 4E (eIF4E), through a conserved binding motif. It′s interaction with eIF4E is a priming event in inducing messenger ribonucleoprotein rearrangement and transition from translation to decay. Eukaryotic initiation factor 4E plays an important role in controlling cell growth. 4E-T is essential for the assembly of processing bodies and its overexpression triggers the movement of eIF4E into the processing bodies.

General description: Eukaryotic translation initiation factor 4E nuclear import factor 1 (eIF4ENIF1) is mapped to human chromosome 22q12.2. It expressed in the human ovary and comprises two leucine-rich nuclear export signals and a bipartite nuclear localization signal.
Immunogen: synthetic peptide corresponding to amino acids 650-664 of human eIF4ENIF1, conjugated to KLH via an N-terminal added cysteine residue. This sequence is specific to human.
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, and 15 mM sodium azide.
Preparation Note: For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
