| Biochem/physiol Actions: | ER degradation-enhancing alpha-mannosidase-like 1 (EDEM1), a putative mannose binding lectin, targets misfolded glycoproteins for degradation in an N-glycan dependent manner. It interacts with the C-terminus of calnexin and lacks mannosidase activity. |
| Biochem/physiol Actions: | ER degradation-enhancing alpha-mannosidase-like 1 is an enzyme encoded by the EDEM1 gene in humans. It is a chaperone for rod opsin and its expression can be used to promote correct folding enhancing degradation of mutant proteins in the ER to combat protein-misfolding disease. The endogenous EDEM1 in cells that are not stressed by the expression of a transgenic misfolded protein reaches the cytosol and is degraded by basal autophagy. EDEM may maintain the retrotranslocation competence of NHK by inhibiting aggregation. It is also considered responsible for directing aberrant proteins for ERAD (ER-associated protein degradation). |
