β-Glucuronidase is routinely used to hydrolyze glucuronide-conjugates of drug metabolites in order to optimize sample preparation prior to LC-MS, GC and ELISA analysis.
LC-MS analysis of codeine-glucuronide enzymatic hydrolysis with six different Sigma-produced β-glucuronidase preparations. The limpet-derived enzyme G8132 performed best with this substrate, with reaction completion within 3–4 hours at 60 °C at 1 unit per mL. Comparative data above was performed at 60 °C at 10 unit per mL.
LC-MS analysis of hydromorphone-glucuronide enzymatic hydrolysis with six different Sigma-produced β-glucuronidase preparations. Molluscan enzymes performed similarly. The reaction was complete within 3–4 hours at 60 °C at 1 unit per mL. The E. coli enzymes exhibited low specificity for this substrate.

Application: β-Glucuronidase from Helix pomatia has been used:
• in the enzymatic treatment of urine samples for liquid chromatography-electrospray ionization-tandem mass spectrometry (LC-ESI-MS/MS) analysis
• in the quercetin extraction procedure from pig or rat tissues for high performance liquid chromatography (HPLC) analysis
• in the enzymatic deconjugation step in urine and plasma sample preparation for gas chromatography-mass spectrometry analysis (GC-MS)

Application: β-glucuronidase was used to hydrolyze conjugated glucuronide and sulfate metabolites.
Biochem/physiol Actions: β-glucuronidase (β-GIc) is an exoglycosidase that catalyzes the breakdown of complex carbohydrates. In humans it converts conjugated bilirubin into the unconjugated form, making bilirubin suitable for reabsorption.

General description: β-Glucuronidase Type H-2 from Helix pomatia is a crude solution of enzymes derived from the Roman snail. Many β-glucuronidases derived from mollusks also contain sulfatase activity.
Other Notes: A further purification of G0876 by gel permeation chromatography.
Other Notes: One Sigma or modified Fishman unit will liberate 1.0 μg of phenolphthalein from phenolphthalein glucuronide per hr at 37 °C at pH 5.0 (30 min assay).
Other Notes: Sulfatase Unit Definition: One unit of sulfatase will hydrolyze 1.0 μmole p-nitrocatechol sulfate per hr at pH 5.0 at 37 °C.
