Immunoblot Whole cell extract of HeLa (lanes A, D) and NIH3T3 (lane B), and recombinant hamster GRP78 protein (Lane C) were separated on SDS-PAGE and blotted with Rabbit Anti-GRP78/BiP (GL-19) (Cat. No. G8918). The antibody was developed with Goat Anti-Rabbit IgG, Peroxidase conjugate (Cat. No. A0545) and a chemiluminescent substrate. Antibody dilution 1:3,000. Lane D: Antibody dilution 1:3,000 + Inhibition with GRP78 immunizing peptide (20 μg/mL).
Enhanced Validation-By Independent Antibodies Immunofluorescence. 3T3 cells were fixed and permeabilized with methanol followed by acetone. Fixed cells were stained with 1:1,000 Anti-GRP78/BiP (GL-19) antibody produced in Rabbit (Cat. No. G8918) (A). The antibody developed using Goat Anti-Rabbit IgG, FITC conjugate antibody (Cat. No. F9887), and 1:1,000 Anti-GRP78/BiP (ET-21) antibody produced in Rabbit (Cat. No. G9043) (B). The antibody was developed using Goat Anti-Rabbit IgG, FITC conjugate antibody (Cat. No. F9887). Results Two Anti-GRP78/BiP antibodies, G8918 (A) and G9043 (B), target different regions of GRP78/BiP show similar staining profiles between the two antibodies, demonstrating Independent Antibody Verification.

Biochem/physiol Actions: GRP78 (78-kDa glucose-regulated protein) is essential for cell viability. GRP78/BiP is induced by stress-inducing agents or by conditions that adversely affect endoplasmic reticulum(ER) function, including oxidative stress, chemical toxicity, Ca2+ ionophores, inhibition of the ER Ca2+ -ATPase, inhibitors of glycosylation and hypoxia. GRP78/BiP is critical for maintenance of Ca2+ homeostasis and the prevention of apoptosis. GRP78/BiP levels are reduced in Alzheimer′s disease brains and the decreased expression of GRP78/BiP is found associated with mis-sense mutations in presenilin-1 (PS-1).
