Enhanced Validation-By Recombinant Expression Antibody recognition: Anti-Huntingtin (N-terminal) antibody produced in Rabbit (Cat. No. H7540) is confirmed by western blot in the cell sample that has been altered to overexpress the target protein-HEK-293T over expressing Huntingtin (Lane 1) while no staining is detected in the wild type sample-HEK-293T (Lane 2). The antibody was developed with Goat Anti-Rabbit IgG, Peroxidase conjugate (Cat. No. A0545) and a chemiluminescent substrate. Antibody dilution: 1:1,000.
Immunoblotting Cell lysates of HEK-293T overexpressing human huntingtin N-terminal fragment (Lane 1,3) and HEK-293T non-transfected (Lane 2), were separated on SDS-PAGE, blotted with Anti-Huntingtin (N-terminal) (Cat. No. H7540) and developed using Goat Anti-Rabbit IgG-Peroxidase (Cat. No. A0545) and chemiluminescent substrate. Lanes 1. Antibody: 0.5 μg/mL 2. Antibody: 0.5 μg/mL 3. Biochem/physiol Actions: Huntingtin (Htt) is essential for embryonic development and neurogenesis. Htt associates with various cell organelles like the nucleus, endoplasmic reticulum and Golgi complex. It is also found in neurites and synapses where it associates with vesicular structures and microtubules, suggesting that it may play a role in intracellular vesicular trafficking, calcium homeostasis, neuronal survival, morphogenesis and transcriptional regulation. Aberrant expansion of glutamine repeats (polyQ) in the N-terminal region of huntingtin (htt) causes Huntington′s disease (HD) which is an autosomal dominant neurodegenerative disorder.
