Immunoblot
Whole cell extract from serum and sodium butyrate-treated NIH3T3 cells was separated on SDS-PAGE and blotted with Anti-Acetyl-Histone H3 [Ac-Lys9] (Cat. No. H9286). The antibodies were developed with Goat Anti-Rabbit IgG-Alkaline phosphatase (Cat. No. A9919) and SIGMA FAST™ BCIP/NBT substrate (Cat. No. B5655). Inhibition with the acetylated [Ac-Lys9] histone H3 peptide (Lane 2) with no inhibition with the non-acetyl-histone H3 peptide (Lane 3).

Immunofluorescence NIH-3T3 Mouse fibroblasts cells were fixed and permeabilized with methanol followed by acetone. Fixed cells were stained with 1:1,000 Anti-acetyl-Histone H3 (Ac-Lys9) antibody produced in Rabbit (Cat. No. H9286) followed by 1:100 Anti-Rabbit IgG (whole molecule)-FITC antibody produced in Goat (Cat. No. F9887). Picture C: cells were counterstained with DAPI (blue) to stain nuclei (Picture A: without DAPI, Picture B: only DAPI).

Biochem/physiol Actions: Histone proteins H3, H4, H2A, and H2B function as building blocks to package eukaryotic DNA into repeating nucleosome units that are folded in higher order chromatin fibers.
Biochem/physiol Actions: In most species, histone H3 is primarily acetylated at lysine 9, 14, 18, and 23. Acetylation at lysine 9 significantly influences histone deposition and chromatin assembly in some organisms. Acetylation of specific lysine residues in H3 is also associated with processes apart from transcription.

• western blot at a dilution of 1:1,000 using whole cell extract of the mouse fibroblast NIH3T3 cell line treated with sodium butyrate
Immunogen: synthetic acetylated [Ac-Lys9] histone H3 peptide (amino acids 7-20) corresponding to the N-terminus of human histone H3. The sequence is identical in many species including mouse, rat, bovine, chicken, frog, Drosophila, and C. elegans, and is highly conserved (single amino acid substitution) in Tetrahymena histone H3.
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
