Enhanced Validation-By Independent Antibodies HeLa cells were fixed and permeabilized with 4% paraformaldehyde followed by 0.5% Triton. Fixed cells were stained with 1:250 Anti-Histone Deacetylase 4 (HDAC4) (ML-19) antibody produced in Rabbit (Cat. No. H9411) (A) and developed using Goat Anti-Rabbit IgG, Cy3 conjugate, and 1:250 Anti-Histone Deacetylase 4 (HDAC4) antibody, Mouse monoclonal, Clone: HADC4-144 (Cat. No. H0163) (B). The antibody was developed using Goat Anti-Mouse IgG, Cy3 conjugate antibody. Results Two Anti-Histone Deacetylase 4 (HDAC4), H9411 (A) And H0163 (B) target different regions of Histone Deacetylase 4 (HDAC4) show similar staining profiles between the two antibodies, demonstrating Independent Antibody Verification.
Immunofluorescence HeLa cells were fixed and permeabilized with 4% paraformaldehyde followed by 0.5% Triton X-100. Fixed cells were stained with 1:250 Anti-Histone Deacetylase 4 (HDAC4) (ML-19) antibody produced in Rabbit (Cat. No. H9411). The antibody was developed using Anti-Rabbit IgG (whole molecule)-FITC antibody produced in Goat (Cat. No. F9887).
Western Blotting Whole extract of 3T3 cells were separated on SDS-PAGE and probed with Anti-Histone Deacetylase 4 (HDAC4) (ML-19) antibody produced in Rabbit (Cat. No. H9411). The antibody was developed using 1:10,000 Anti-Rabbit IgG (whole molecule)-Peroxidase antibody produced in Goat (Cat. No. A0545). Lanes 1. 1:1,000 antibody 2. 1:2,000 antibody 3. Negative Control

Biochem/physiol Actions: Histone deacetylase 4 (HDAC4) exhibits intrinsic nuclear import and export signals for its dynamic nucleocytoplasmic shuttling. Its association with 14-3-3 and monocyte enhancer factor 2 (MEF2) proteins can disturb shuttling and thus directs HDAC4 to the cytoplasm and the nucleus, respectively.

General description: Mammalian histone deacetylases (HDACs) can be divided into three classes according to sequence homology. Class II consists of the yeast Hda1-like proteins histone deacetylase 4,5,6,7,8,9, 10 (HDAC4, HDAC5, HDAC6, HDAC7, HDAC9, and HDAC10.
Immunogen: synthetic peptide corresponding to amino acid residues 1-19 of human HDAC4 with C-terminal added lysine conjugated to KLH.
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 1% bovine serum albumin and 15 mM sodium azide.
