| Biochem/physiol Actions: | RNF139 (ring finger protein 139) gene encodes a endoplasmic reticulum protein that contains a RING-H2 finger required for Ub ligase activity and a sterol-sensing domain. It possesses E3-ubiquitin ligase activity. It negatively regulates cell proliferation via mechanisms involving G2/M arrest, decreased DNA synthesis and cell death. It acts as a putative tumor suppressor. It represses genes involved in the biosynthesis of cholesterol and fatty acids that are transcriptionally regulated by the sterol response element binding proteins (SREBPs), thus, establishing a link between growth control and the cholesterol/lipid homeostasis pathway. It inhibits the transport of sterol regulatory element-binding protein-2 (SREBP-2) from ER to Golgi. It is linked to the development of hereditary kidney cancer. The protein is required for the dislocation of MHC I (major histocompatibility complex class I) from the ER. |
