| Biochem/physiol Actions: | 15-hydroxyprostaglandin dehydrogenase (HPGD) has a functional colon cancer tumor suppressor activity. This enzyme catalyzes the inactivating NAD+-dependent conversion of the prostaglandin 15-OH group to a 15-keto group. Its expression is directly controlled and strongly induced by activation of the TGF-β tumor suppressor pathway. The enzymatic pathway that induces colon cancer suppression is activated by TGF-β and mediated by HPGD. It also catalyzes the NAD-dependent dehydrogenation of lipoxin A4 (LXA4) to form 15-oxo-lipoxin A4 by the dehydrogenation of the C15 hydroxyl group of LXA4 to an oxo- group hence forming 15-oxo-LXA4. Mutations in this gene can cause primary hypertrophic osteoarthropathy and cranioosteoarthropathy. |
