| Biochem/physiol Actions: | HGS (hepatocyte growth factor-regulated tyrosine kinase substrate) gene encodes a FYVE domain protein localized to early endosomes. It functions in membrane trafficking of various molecules. It recruits Smad2 and Smad3 to the activin receptor complex in cooperation with SARA and regulates Smad signaling involved in mouse early embryonic development. It functions in the regulation of endosomal sorting and targets proteins for lysosome-dependent degradation. Rapid phosphorylation of Hgs is stimulated by IL-2, granulocyte-macrophage colony-stimulating factor and hepatocyte growth factor. It is associated with STAM, a signal-transducing adaptor molecule, and suppresses its function in cell growth signaling mediated by the cytokines. |
