| Biochem/physiol Actions: | Nuclear factor NF-κ-B p100 subunit (NFKB2) is at the endpoint of many of signal transduction pathways which are initiated by stimuli such as inflammation, immunity, differentiation, cell growth, tumorigenesis and apoptosis. It forms a homo- or heterodimeric complex with proteins. These complexes bind at κ-B sites in the DNA of their target genes. NFKB2 complexes are present in the cytoplasm in an inactive state bound to the NF-κ-B inhibitors. For the activation of NFKB2, I-κ-B (inhibitor) is phosphorylated by I-κ-B kinases (IKKs) and degraded. Thus the active NF-κ-B complex is free from the inhibitor and can translocate to the nucleus. NFKB2 plays an important role in the cytoplasmic retention of attached NFKB2 proteins by p100 and the generation of p52 by cotranslational processing. It also regulates the circadian clock by repressing the transcriptional activator activity of the complex formed by aryl hydrocarbon receptor nuclear translocator-like protein 1 (ARNTL) with other proteins. |
