| Biochem/physiol Actions: | SAMSN1 (SAM domain, SH3 domain and nuclear localization signals 1) protein may be involved in the formation of blood vessel during normal brain development. It may function as an adaptor or scaffolding protein. It has been found to be overexpressed in glioma and is associated with a poor survival in glioblastoma multiforme. In mice, this protein may function in moderating adaptive immune responses. SAMSN1 may be involved in B cell activation and differentiation. It has been suggested to be a tumor suppressor in lung cancer. It may serve as a biomarker of the phenotype of human hepatocellular carcinoma. |

| General description: | SAMSN1 (SAM domain, SH3 domain and nuclear localization signals 1) gene encodes a member of the SLY family of cytoplasmic adaptor proteins. It is abundantly expressed in the hematopoietic compartment. The protein is also expressed in heart, brain, placenta, and lung to a lesser extent. The members of this family are characterized by the presence of protein-protein interaction domains and are homologous. They contain a sterile α motif (SAM) domain that is capable of self-association and can also associate with each other. The chimeric fusion of this domain with the βPDGF receptor, AML1, c-Abl, and JAK2 results in the oncogenic transformation of the SAM domain. The SH3 domains mediate protein-protein interactions in several signal transduction pathways. The gene is localized to human chromosome 21 (21q11.2). |
