| Biochem/physiol Actions: | GCNT3 (glucosaminyl (N-acetyl) transferase 3, mucin type) is responsible for the production of the three β6GlcNAc branches on secreted mucins. Not only does it catalyze the synthesis of core 2 branches, it also catalyzes the synthesis of core 4 and 1 branches. Its expression is suppressed by epidermal growth factor, and enhanced by all-trans retinoic acid and T helper 2 (Th2) cytokines such as IL-13. These cis-regulatory elements, therefore, have potential as therapeutic targets for disorders with mucus hyper-secretion. GCNT3 is responsible for the modification of O-glycans and integrin signaling, which in turn affect cell adhesion and migration and tumorigenesis. This gene is under-expressed in majority of colorectal cancer patients. It has potential to determine the relapse risk in early stage colon cancer patients, and might also help to determine the response to chemotherapy in the same. |
