| Biochem/physiol Actions: | Peptidyl-tRNA hydrolase 2 (PTRH2) takes part in integrin-mediated cell survival and apoptosis signaling. PTRH2 prevents the peptidyl-tRNAs from accumulating once they dissociate from the ribosomes. After the integrin-mediated cell attachment, it interacts with focal adhesion kinase (FAK) and induces cell survival. It has an important role in anoikis, cell death due to loss of cell attachment to the extra cellular matrix. When this occurs, PTRH2 moves from mitochondria into the cytosol. It then forms a complex with the transcriptional regulator amino-terminal enhancer of split (AES) to mediate apoptosis. PTRH2 suppresses the phosphorylation of extracellular signal-regulated kinase (ERK). It regulates nuclear factor κβ (NFκβ) and ERK signaling. Mutations in the gene have been associated with an infantile multisystem neurologic, endocrine and pancreatic disease (IMNEPD). |
