Biochem/physiol Actions: Prostacyclin synthase encoded by the gene PTGIS catalyzes the conversion of PGH2 (Prostaglandin H2) to PGI2 (prostaglandin I2). PGH2 is derived from arachidonic acid by COX1 (cytochrome c oxidase subunit 1) and COX2. PGI2 is found to have antiproliferative effects and chemopreventive properties. Intracellular PGI2 activates endogenous PPARδ (peroxisome proliferator activated receptor δ) receptor and triggers apoptosis. Epigenetic inactivation of PTGIS gene has been found frequently in colorectal carcinogenesis.

General description: The gene PTGIS (prostaglandin I2 synthase) encodes a protein belonging to the cytochrome P450 superfamily of enzymes. The encoded protein is also referred to as prostacyclin synthase. The gene is made up of 10 exons spanning a length of approximately 60kb. It is mapped to human chromosome 20q13.11–q13.13. The 5′ flanking section of the gene contains the core binding sequence (GAGACC) of the shear stress responsive element. Two different mRNA transcripts have been found in human aorta and lung, one 6kb and the other spanning a length of 3.3kb.
