Biochem/physiol Actions: C5AR1 (complement component 5a receptor 1) binds to C5a, which results in G-protein dependant cellular activation. This includes rise in intracellular Ca2+ concentration and activation of mitogen-activated protein kinase (MAPK), PKB/Akt and phosphoinositide 3-kinase pathways. C5a is an anaphylatoxin, which has potent pro-inflammatory actions, and is involved in the pathogenesis of asthma, rheumatoid arthritis and sepsis. C5AR is involved in immunity against pathogens such as, bacteria like P. aeruginosa. It initiates their phagocytosis and degradation. It is expressed on Müller cells, which are glial cells found in eye. C5AR might be involved in the pathogenesis of retinal diseases such as, diabetic retinopathy (DR), by inducing the production of IL (interleukin)-6 and VEGF (vascular endothelial growth factor).

General description: C5AR1 (complement component 5a receptor 1) is a G-protein coupled receptor (GPCR) belonging to the rhodopsin family. It is expressed in myeloid-lineage blood cells, as well as non-myeloid cells such as epithelial cells of bronchi and alveolus, and astrocytes. Its ligand is C5a, and binds to it through two sites of ligand-binding domain. One site binds to the C-terminal of C5a and the other site is a part of the N-terminal of C5AR1. C5AR1 along with two more receptors, C3AR and C5L2, make up the anaphylatoxin (AT) receptor family of the complement system.
