Enhanced Validation-By Independent Antibodies Immunohistochemical staining of human kidney, liver, lymph node and small intestine using Anti-SP100 antibody HPA016707 (A) shows similar protein distribution across tissues to independent antibody HPA017384 (B).
Enhanced Validation-Orthogonal RNAseq Confirmation Western blot analysis in human cell lines SK-MEL-30 and HEK293 using Anti-SP100 antibody. Corresponding SP100 RNA-seq data are presented for the same cell lines. Loading control: Anti-PFN1.
Immunohistochemistry Immunohistochemical staining of human lymph node shows strong nuclear positivity in non-germinal center cells.
Immunohistochemistry Immunohistochemical staining of human small intestine shows strong nuclear positivity in glandular cells.
Immunohistochemistry Immunohistochemical staining of human kidney shows moderate nuclear positivity in cells in tubules.
Immunohistochemistry Immunohistochemical staining of human liver shows no positivity in hepatocytes as expected.

Application: Anti-SP100 antibody produced in rabbit, a Prestige Antibody, is developed and validated by the Human Protein Atlas (HPA) project  . Each antibody is tested by immunohistochemistry against hundreds of normal and disease tissues. These images can be viewed on the Human Protein Atlas (HPA) site by clicking on the Image Gallery link. The antibodies are also tested using immunofluorescence and western blotting. To view these protocols  and other useful information about Prestige Antibodies and the HPA, visit sigma.com/prestige .
Application: Applications in which this antibody has been used successfully, and the associated peer-reviewed papers, are given below.

Biochem/physiol Actions: SP100, a cellular protein, is a major component of the nuclear domain 10 (ND10). It encodes a major component of the PML (promyelocytic leukemia) SP100 nuclear bodies, which plays a vital role in viral infection control. During transcriptional regulation, it acts as a transcriptional and chromatin regulatory factor.

Features and Benefits: Prestige Antibodies® are highly characterized and extensively validated antibodies with the added benefit of all available characterization data for each target being accessible via the Human Protein Atlas portal linked just below the product name at the top of this page. The uniqueness and low cross-reactivity of the Prestige Antibodies® to other proteins are due to a thorough selection of antigen regions, affinity purification, and stringent selection. Prestige antigen controls are available for every corresponding Prestige Antibody and can be found in the linkage section.

Every Prestige Antibody is tested in the following ways:
• IHC tissue array of 44 normal human tissues and 20 of the most common cancer type tissues.
• Protein array of 364 human recombinant protein fragments.
Immunogen: Nuclear autoantigen Sp-100 recombinant protein epitope signature tag (PrEST)
