| Biochem/physiol Actions: | Ancient conserved domain-containing protein 4 (CNNM4) is involved in metal ion transport. It may remove unwanted Mg2+ ions which interfere with crystal formation in enamel and thus maintain the serum Mg2+ levels. In the epithelium, CNNM4 expels intracellular Mg2+ outside the cells in exchange for Na+. It binds to the enzyme phosphatase of regenerating liver (PRL) which is up-regulated in malignant human cancers. Along with PRL, CNNM4 is involved in energy metabolism and 5′ AMP-activated protein kinase (AMPK)/ mechanistic target of rapamycin (mTOR) signaling. Mutations in CNNM4 have been shown to be associated with recessive cone-rod dystrophy and amelogenesis imperfecta (Jalili syndrome). |

| General description: | Ancient conserved domain-containing protein 4 (CNNM4) contains a highly conserved domain which is evolutionarily conserved from bacteria to mammals. In total, it has four transmembrane domains. CNNM4 is expressed in the gastrointestinal tract, neural retina and ameloblasts of the developing tooth. The gene encoding it is present on chromosome 2q11. |
