| Biochem/physiol Actions: | In the biogenesis of iron-sulfur proteins, the final step of iron-sulfur protein assembly involves transfer of an iron-sulfur cluster from a cluster-donor to a cluster-acceptor protein. This process is mediated by a molecular chaperone from the Hsc70 (Heat shock cognate) family and a co-chaperone of the J-domain family. HSCB (Iron-sulfur cluster co-chaperone protein HscB, mitochondrial) is homolog of DnaJ type co-chaperones. It is required for mitochondrial iron-sulfur cluster (ISC) assembly. Depletion of HSCB reduces mitochondrial and cytosolic ISC-containing enzymes. On the other hand, overexpression of HSCB protects cells from oxidative stress. HSCB interacts with iron-sulfur cluster scaffold protein and mitochondrial HSP70 (Heat shock protein). |

| General description: | The gene HSCB (iron-sulfur cluster co-chaperone protein HscB, mitochondrial) is mapped to human chromosome 22q12.1. It is expressed in various tissues and is mainly present in the mitochondria. However, small amount of HSCB is also detected outside mitochondria. It contains a conserved amino-terminal J-domain and a less conserved carboxyl-terminal region. It is popularly known as HSC20 (Heat shock cognate 20). |
