| Biochem/physiol Actions: | VPS9D1 (Vacuolar Protein Sorting 9 Domain-containing gene 1) gene encodes a protein containing VPS9 domain with ATP synthase and GTPase activator activity. The VPS9 domain is highly conserved and is involved in the activation of Rab5 GTPase, which mediates cell signaling via endocytosis of intracellular receptors. It also associates with GRB2 (growth factor receptor-bound factor 2), an adaptor protein that binds SOS1 (SOS Ras/Rac guanine nucleotide exchange factor 1) in response to growth factors, and activates membrane-bound Ras, resulting in increased cell proliferation and survival. GRB2 is also involved in calcium-regulated signaling in B-cells. Together, the increased expression of VPS9D1 and its missense variants along with upregulation of GRB2 that is observed in survivors of sepsis, results in enhanced cellular proliferation and survival, activation of B- and T-cells, and proapoptotic activity. |
