Biochem/physiol Actions: Chromosomal alteration at der(17)t(X:17)(p11:q25) leads to the fusion of alveolar soft part sarcoma chromosomal region candidate gene 1 (ASPSCR1) and transcription factor E3 (TFE3) to form the complex ASPSCR1–TFE3 in paraffin-embedded alveolar soft part sarcomas (ASPS).This fusion transcript plays a vital role in differential diagnosis of ASPS. TFE3 forms a complex with ASPL by replacing its N-terminal portion with fused ASPL sequences, but TFE3 retains its DNA-binding domain facilitating transcriptional deregulation in the pathogenesis of ASPS. The protein encoded by ASPSCR1 consists of conserved domains and it might be involved in the ubiquitylation pathway,but specific function of this protein remains unknown. In 3T3-L1 (cell line derived from mouse 3T3 cells) adipocytes, ASPSCR1 controls the trafficking of glucose transporter type 4 (GLUT 4). The encoded protein initiates p97 hexamer disassembly and it is also involved in the reformation of Golgi complex after brefeldin A (BFA) removal.

General description: Alveolar soft part sarcoma chromosomal region candidate 1 (ASPSCR1) gene is mapped to human chromosome 17q25. ASPSCR1, also known as alveolar soft part sarcoma locus (ASPL) or TUG (tether, containing a UBX domain, for GLUT4) codes for a member of UBX – domain containing proteins. The encoded protein consist of 553 amino acids and it is characterized with a C-terminal ubiquitin regulatory X (UBX)-like domain. ASPSCR1 is ubiquitously expressed in all adult tissues.
