| Biochem/physiol Actions: | O-GlcNAc (O-linked N-acetylglucosamine) transferase (OGT) catalyzes O-GlcNAcylation that occurs in mitochondria, nuclear, and cytoplasmic proteins. O-GlcNAc transferase is a subunit of nonspecific lethal complex, a transcriptional regulator. O-GlcNAcylation involves the addition of O-GlcNAc to specific serine or threonine residue on many of the cytosolic and nuclear proteins. Disturbance in O-GlcNAcylation is associated with the development of diseases including cancer, diabetes, and Alzheimer disease. OGT is associated with increased mitochondrial respiration and elevated glycolysis. Inhibition in addition or removal of O-GlcNAc leads to a defect in preinitiation complex assembly. |

| General description: | OGT (O-linked N-acetylglucosamine transferase) gene is mapped to human chromosome Xq13.1. This gene encodes for nucleo cytosolic and mitochondrial isoforms of the enzyme. OGT is highly conserved among Caenorhabditis elegans, rat, and human genomes, sharing over 65% amino acid identity. The gene includes multiple tandem tetratricopeptide repeat sequences and is modified by O-GlcNA (O-linked N-acetylglucosamine), also by tyrosine phosphorylation. |
