| Biochem/physiol Actions: | IDH1 (isocitrate dehydrogenase (NADP) 1 cytosolic) participates in the TCA (tricarboxylic acid) cycle. It is responsible for the formation of α-ketoglutarate (αKG) by oxidative decarboxylation of isocitrate, thereby resulting in the generation of NADPH (nicotinamide adenine dinucleotide phosphate). IDH1 helps in glucose sensing, glutamine metabolism, lipogenesis and controlling cellular redox condition. In presence of hypoxia, it results in reductive carboxylation of αKG to form acetyl-CoA, which is needed for lipid synthesis. In mouse model, overexpression of the IDH1 gene causes obesity, fatty liver and hyperlipidemia. Mutations in this gene are associated with glioblastoma multiforme and acute myeloid leukemia. |
