ZFYVE27 gene mutation leads to an autosomal dominant form of hereditary spastic paraplegia (AD-HSP). The encoded membrane protein regulates polarized vesicular trafficking in neurons. ZFYVE27 and spastin protein complex play an essential role in motor axon outgrowth. |

| General description: | Zinc finger FYVE-type containing 27 (ZFYVE27), also called as SPG33 or protrudin, is encoded by the gene mapped to human chromosome 10q24.2. ZFYVE27 is a specific spastin-binding protein and is a member of Fab1, YOTB/ZK632.12, Vac1, and EEA1 (FYVE)-finger family of proteins.The encoded protein contains N-terminal Rab11 binding domain (RBD11) and C-terminal FYVE domain. In addition, it also contains two phenylalanines (FF) in an acidic tract (FFAT) motif, a coiled-coil domain and three hydrophobic region (HR) motifs in the central region of the protein. |
