Biochem/physiol Actions: Heat shock protein family A (Hsp70) member 5 (HSPA5) facilitates embryonic cell growth and pluripotent cell survival. In addition, it also assists various cellular processes such as protein transport into the endoplasmic reticulum (ER), translocation of nascent polypeptides, de novo protein folding and assembly and ER associated degradation (ERAD). It is also involved in regulation of calcium homeostasis. HSPA5 is linked to stress-associated diseases. The gene also promotes progression of various critical human diseases, including cancers, viral infections and neurodegenerative disorders. Polymorphisms in this gene might function as biomarkers to determine the risk and poor prognosis of neuroblastoma (NB).

General description: Heat shock protein family A (Hsp70) member 5 (HSPA5), also known as glucose/regulated protein 78 (GRP78), is a stress-associated protein, encoded by the gene mapped to the long arm of human chromosome 9q33.3. The encoded protein belongs to the heat shock protein seventy family and is expressed primarily in the endoplasmic reticulum (ER). HSPA5 is a 654 amino acid protein, characterized by an N-terminal nucleotide binding domain with ATPase activity, a substrate binding domain and a variable C-terminal tail.
