Biochem/physiol Actions: Inositol hexakisphosphate kinase 1 (IP6K1) is involved in the synthesis of diphosphoinositol pentakisphosphate. It interacts with cullin–RING ubiquitin ligase-4 (CRL4) and has a role in DNA homologous recombination repair. In addition, IP6K1 is also implicated in chromatin modifications and spermatogenesis. Elevated expression or mutation of IP6K1 is associated with the pathogenesis of type 2 diabetes mellitus and crohn′s disease. IP6K1 plays a vital role in dynein-dependent trafficking pathways, including endosomal sorting, vesicle movement and Golgi maintenance. IP6K1 controls DNA repair and cell death by mediating disassembly of the cullin-RING ubiquitin ligase 4 (CRL4)-COP9 signalosome (CSN) complex.
