| Application: | Monoclonal Anti-Interleukin-6 antibody may be used for ELISA at a minimum antibody concentration of 4 μg/ml. For immunoblotting a working antibody concentration of 1-2 μg/ml may be used. The antibody may be used in neutralization assays; ND50 is 0.01-0.15 μg/ml. For neutralization of IL-6 from human peripheral blood mononuclear cells, a concentration of 2.5 μg/ml was used. |

| General description: | Interleukin-6 is a pleotropic cytokine involved in hematopoiesis, inflammation and immune response. IL-6 is secreted by macrophages, dendritic cells and B cells, in response to TNF-α, PGDF and IL-1. The signaling by IL-6 is mediated by heterodimeric receptor, IL-6R, comprising of IL-6Rα (soluble factor) and gp130 (sometimes called IL-6Rβ). gp130 is crucial component for downstream signal transduction. In response to IL-6, gp130 activates kinases of JAK family, STATs, MAPK and Ras proteins. IL-6-mediated signaling is essential for normal development of T and B cells and has been implicated in many diseases such as rheumatoid arthritis, Crohn disease, sepsis, fever, systemic lupus erythematosus, osteoporosis and insulin resistance. Monoclonal Anti-Interleukin-6 antibody recognizes human IL-6. The antibody shows no cross-reactivity with recombinant mouse IL-6 or recombinant human IL-6 sR, OSM, LIF CNTF, or IL-11 or recombinant horse gp130. |
