Immunofluorescence
HeLa cells were fixed and permeabilized with 3% paraformaldehyde followed by 0.5% Triton-X-100. Fixed cells were stained with 1 μg/mL Anti-Lamin A (C-terminal) (Cat. No. L1293) and developed with Goat Anti-Rabbit IgG, Cy3 conjugate.
Enhanced Validation-By Independent Antibodies HeLa cells were fixed and permeabilized with 4% paraformaldehyde followed by 0.5% Triton. Fixed cells were stained with 0.5 μg/mL Anti-Lamin A, mature antibody, Mouse monoclonal, Clone: 4A4 (Cat. No. SAB4200420) (A) and developed using Goat Anti-Mouse IgG, Cy3 conjugate, and 1 μg/mL Anti-Lamin A (C-terminal) antibody produced in Rabbit (Cat. No. L1293) (B). The antibody was developed using Goat Anti-Rabbit IgG, FITC conjugate antibody. Results Two Anti-Lamin A antibodies, SAB4200420 (A) and L1293 (B) target different regions of Lamin A show similar staining profiles between the two antibodies, demonstrating Independent Antibody Verification.
Immunofluorescence NIH/3T3 Mouse fibroblast cells were fixed and permeabilized with 4% paraformaldehyde followed by 0.5% Triton X-100. Fixed cells were stained with 2 μg/mL Anti-Lamin A (C-terminal) antibody produced in Rabbit (Cat. No. L1293). The antibody was developed using Anti-Rabbit IgG (whole molecule)-FITC antibody produced in Goat (Cat. No.F9887).
Immunofluorescence NRK normal Rat kidney cells were fixed and permeabilized with 4% paraformaldehyde followed by 0.5% Triton X-100. Fixed cells were stained with 2 μg/mL Anti-Lamin A (C-terminal) antibody produced in Rabbit (Cat. No. L1293). The antibody was developed using Anti-Rabbit IgG (whole molecule)-FITC antibody produced in Goat (Cat. No.F9887).
Western Blotting HeLa cells nuclear extract were separated on SDS-PAGE and probed with Anti-Lamin A (C-terminal) antibody produced in Rabbit (Cat. No. L1293). The antibody was developed using 1:30,000 Anti-Rabbit IgG (whole molecule)-Peroxidase antibody produced in Goat (Cat. No. A0545). Application: Anti-Lamin A (C-terminal) antibody is suitable for use in western blot (0.1-0.2 μg/mL) using HeLa nuclear extract. This antibody can also be used in indirect immunofluorescence (1-2 μg/mL) using HeLa cells, rat NRK and mouse 3T3 cells. Additionally, anti-Lamin A (C-terminal) antibodies are suitable for use in immunoblotting (approx. 70 kDa). Cleaved fragments of lamin A may form additional bands at 45-50 kDa.
Application: Anti-Lamin A (C-terminal) antibody produced in rabbit has been used in:

General description: Lamin is a structural protein of the nuclear lamina. Lamin A is a type A lamin encoded by the LMNA gene. Lamin A contains 664 amino acids and is expressed in most somatic cells. It contains α-helical rod domain to enable assembly into filaments, a nuclear localization sequence, and a carboxy-terminal CAAX box isoprenylation sequence for nuclear membrane targeting.
Immunogen: synthetic peptide corresponding to amino acids 598-611 of human lamin A with a C-terminal added cysteine, conjugated to KLH. The corresponding sequence differs by one amino acid in rat, three amino acids in mouse, and by a gap of one amino acid in both rat and mouse lamin A.
Physical form: Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
