Immunofluorescence NRK Rat kidney cells were fixed and permeabilized with 4% paraformaldehyde followed by 0.5% Triton X-100. Fixed cells were stained with1:100 Anti-Myosin IIA, non-muscle antibody produced in Rabbit (Cat. No. M8064). The antibody was developed using Anti-Rabbit IgG (whole molecule)-FITC antibody produced in Goat (Cat. No. F9887). Cells were counterstained with DAPI (blue) to stain nuclei.
Western Blotting Whole extract of MDCK cells were separated on SDS-PAGE and probed with Anti-Myosin IIA, non muscle antibody produced in Rabbit (Cat. No. M8064). The antibody was developed using 1:5,000 Anti-Rabbit IgG (whole molecule)-Alkaline Phosphatase antibody produced in Goat (Cat. No. A9919). Biochem/physiol Actions: Nonmuscle myosin II is involved in cell motility and adhesion, cytokinesis, vesicular transport, intracellular force generation and in morphogenesis during development. Its activity is regulated by light chain and possibly heavy chain phosphorylation and by association with proteins such as Mts1. Mutations in the NMHCA gene are found in several syndromes associated with megakaryocyte/platelet/leukocyte disorders. Mutations in the MYH9 gene causes a spectrum of macrothrombocytopenia disorders with neutrophil inclusions, termed as MYH9 disorders.
